研 究 興 趣
過去數年來,我們對正常肝臟與肝癌組織之基因表現深感興趣,本實驗室應用先進科技,大量分析基因之表現。研究結果顯示蛋白激脢在不同生理病理狀況下有不同的表現型式。我們也透過團隊合作,建立研究肝臟相關研究所需之基因庫材料並且分析了數千筆肝臟之基因序列。這些資訊可供研究者參考,從中找尋研究的新方向。最近,我們更致力建立百萬鹼基級之基因定序能力,除了協助搜尋可能之抑癌基因外,亦可對人類基因組計劃有所貢獻。

人類基因體計畫 (The Human Genome Project),自1998年進入一個高峰。藉由先前遺傳圖譜 (genetic map)及物理圖譜 (physical map) 之基礎,全球之基因體研究重點轉向人類基因體之定序工作。目前已經發表有完整之第二十一、二十二號兩個染色體之完成序列 (finished sequence),以及其它染色體之初稿序列 (draft sequence),這些基因體資訊提供未來生物醫學研究之重要基礎。本人實驗室自1998年開始規劃台灣之大規模基因體定序能力,建立年產千萬鹼基 (10 Mb)之核心設施,截至目前已對公眾資料庫提供1200萬以上高精確度之序列,主要集中於人類第四號染色體4q21-24 區域,此區域在肝癌病患肝臟細胞基因體經常出現缺失。這些成果可參考網頁資料(網址 http://genome.ym.edu.tw)。

在4q21-24 區域中,除了可能之抑癌基因 (tumor suppressor gene) 外,尚有上百個基因,其中有些與人類之疾病有關。例如 PKD2基因與體染色體顯性之成年型多囊腎疾病(polycystic kidney disease)有關,SNCA 基因與巴金森氏症(Parkinson disease)有關。這些基因具有重要之醫學意義,值得進一步探討。由於基因表現調控受到染色體局部及整體環境之影響,人類基因體序列除了可以協助發現基因(gene discovery)外,亦對研究基因表現之調控機制提供新的線索。本實驗室完成人類ADH 基因與小鼠Adh基因之比較分析,找出控制整個第一型ADH基因座(Class I ADH gene complex)之關鍵區域。目前正以轉殖鼠技術與生化學方法進一步研究基因調控之機制。

基因體科技之進展逐漸與臨床醫學結合,對於疾病之診斷、分類、治療與預防作出貢獻。本人實驗室已建立基因體分析技術,例如微陣列及突變分析之標準程序,將可找出與疾病相關之基因,並進一步剖析基因變異造成人類疾病之病理過程。這些資訊的取得雖然費時耗力,但將可加速對人類疾病之掌握,甚至帶動對於病人治療方式之新的突破。

由於基因的運作原理是演化保留下的生物基本法則,本實驗室運用此一觀念找尋人類與果蠅共通之基因並研究其功能。目前已發現數個基因與調節染色質結構及基因表現有關,目前正進一步探討其生理功能。
